Arginase is a manganese-containing enzyme performing the chemical change of l-arginine to urea and ornithine. Because of arginine’s role as a central metabolic player, arginase has become a primary ongoing research target with multiple roles in human health and disease. In this chapter, I discuss the structural details of the enzyme, and how structural biology is shaping the development of arginase small-molecule inhibitors and inhibitor-antibodies. These studies represent the foundation of arginase for therapeutic strategies in many areas of medicine ranging from immune response and aging to oncology.

Abstract

Arginase is a manganese-containing enzyme performing the chemical change of l-arginine to urea and ornithine. Because of arginine’s role as a central metabolic player, arginase has become a primary ongoing research target with multiple roles in human health and disease.
