The interaction of the porphyrin derivative H2TCPPSpm4, having four spermines in the meso positions, with the G-quadruplex (GQ) structure formed by the DNA aptamer TGGGAG, a sequence that show anti-HIV activity, has been investigated by means of UV, CD and PAGE studies. The results demonstrate that porphyrin substituted with positively charged side chains is capable of inducing the formation of different complexes with parallel GQs, including higherordered complexes, depending on the method used to achieve their relative stoichiometry (titration vs single addition). Indeed the CD melting experiments indicated different thermal stability fort the 1:1 H2TCPPSpm4:GQ and the 2:1 H2TCPPSpm4:GQ species obtained by titration. Interestingly also the 2:1 H2TCPPSpm4:GQ complexes obtained by titration or by direct mixing show different behavior.
