Onconase is a member of the pancreatic ribonuclease A (RNase A) superfamily, initially it was isolated from extract of Rana pipiens oocytes and early embryos. Onconase has cytotoxic effects toward tumor cells. Onconase acquires more cytotoxic effect if it is conjugated with cell-type specific ligands and it has special properties such as anti-microbial activity. In this study we characterize the antimicrobial activity of wild type onconase, and different mutants against Gram negative and positive bacteria, and fungi. Using bio-informatic programs we obtained a variety of onconase mutants, in some of these we have added positive charges to the protein, in others we have increased the onconase sensitivity to bacterial proteases.

Abstract
Onconase is a member of the pancreatic ribonuclease A (RNase A) superfamily, initially it was isolated from extract of Rana pipiens oocytes and early embryos. Onconase has cytotoxic effects toward tumor cells. Onconase acquires more cytotoxic effect if it is conjugated with cell-type specific ligands and it has special properties such as anti-microbial activity. In this study we characterize the antimicrobial activity of wild type onconase, and different mutants against Gram negative and positive bacteria, and fungi. Using bio-informatic programs we obtained a variety of onconase mutants, in some of these we have added positive charges to the protein, in others we have increased the onconase sensitivity to bacterial proteases.I documenti in IRIS sono protetti da copyright e tutti i diritti sono riservati, salvo diversa indicazione.
