The formation of fusicoccin (I) by Fusicoccum amygdali was investigated by feeding cultures with DL-mevalonic-2-14C acid labeled with 3H in either the 2, 4, or 5 positions. It was shown that 1 of the four 4-pro-R mevalonic H atoms retained in I is located on C-15. Thus, whereas the 1st cyclization step of the tetraprenyl precursor of fusicoccins is analogous to that of the pentaprenyl precursor of orphiobolins, the further transformation of (II) is totally different. The H atoms at C-12 and C-13 of I originated from the C-2 and C-5 mevalonoid H atoms, resp. Schemes for I formation and metab. are presented.
