Eight suppression subtractive libraries, enriched in transcripts differentially expressed in response to drought stress in durum wheat were produced and used as source of EST clones. The libraries were made from RNA isolated at different developmental stages (from seedling to maturity) and from different tissues. About 11,000 electropherograms were analysed, leading to 8,457 high quality EST submissions. Then, over-represented ESTs were identified in order to estimate the level of redundancy in each library as well as in the whole EST datasets, and a total of 4,748 non-redundant ESTs were obtained. These sequences significantly contribute to the publicly available EST collection of durum wheat, very limited compared to the corresponding collection of bread wheat, and represent a valuable tool for a better understanding of the mechanisms governing stress tolerance in durum wheat.
